Effects of Pyridazinone Herbicides during Chloroplast Development in Detached Barley Leaves II. Effects on Lipid Content, Fatty Acid Composition and Ultrastructure of Chloroplasts

چکیده
The effects o f three differently substituted pyridazinone herbicides were studied on the lipid content, fatty acid composition o f glycerolipids, fatty acid synthesizing capacity and ultrastruc ture of chloroplasts after 72 h o f greening o f etiolated barley leaves. SAN 9789 caused a nearly threefold increase in the total fatty acid content o f chloroplasts, and this increase was accom panied by an increase in the D G D G and phospholipid-content as well. SAN 6706 and 9785 caused reduction in the linolenic to linoleic acid ratio in nearly all lipid classes, only PC was unaffected. Both herbicides reduced the saturated to unsaturated ratio in the PG, SAN 9789 had no effect on the fatty acid com position o f the PG. Fatty acid synthesis was inhibited in the SAN 6706and 9789-treated leaves as monitored by incorporation o f labelled acetate, but all o f the three pyridazinones stimulated the incorporation o f labelled galactose into chloroplast galactolipids. SAN 6706 and 9789 also caused abnormal organization o f chloroplast lamellar system.

Pyridazinone Herbicides, Carotenoids, Fluorescence Spectrum The effects o f three differently substituted pyridazinone herbicides on the accumulation o f photosynthetic pigments and on the fluorescence properties o f greening barley leaves were studied. The photosynthetic activities of barley leaves in vivo and o f thylakoids, isolated from the leaves, in vitro were studied during greening in the presence or absence of 2 x 10-4 m SAN 6706. The degree o f chlorophyll bleaching increased from 32% at 24 h o f greening to 60% at the final stage. Treated leaves were unable to carry out 14C 0 2-fixation, their fluorescence induction properties showe...
متن کاملInteractions of lipoidal materials and a pyridazinone inhibitor of chloroplast development.
Formation of chloroplast pigments was inhibited, and free fatty acids accumulated in mustard (Brassica juncea [L.] Coss.) cotyledons and in barley (Hordeum vulgare L.) first leaves developed after treatment with 4-chloro-5- (dimethylamino)-2- (alpha, alpha, alpha-trifluoro-m-tolyl) -3 (2H) -pyridazinone. The inhibitor reduced the amount of fatty acids found in polar lipids (galactolipids) of ba...
متن کاملCytokinin stimulates chloroplast transcription in detached barley leaves.
Chloroplasts are among the main targets of cytokinin action in the plant cell. We report here on the activation of transcription by cytokinin as detected by run-on assays with chloroplasts isolated from apical parts of first leaves detached from 9-d-old barley (Hordeum vulgare) seedlings and incubated for 3 h on a 2.2 x 10(-5) m solution of benzyladenine (BA). Northern-blot analysis also detect...
متن کاملPolar lipid composition of a plastid ribosome-deficient barley mutant.
Green and white leaves of the barley mutant line ;albostrians' were compared for their polar lipid content and fatty acid composition. The mutant plastids of the white leaves have a double-layered envelope, but in contrast with the normal chloroplasts, lack 70 S ribosomes and thylakoids. In the green leaves, the amount of monogalactosyldiacylglycerol (MGDG) consistently exceeds the amount of di...
